چکیده

Sir, We read with great interest the article on “Comparison of potassium hydroxide mount and mycological culture with histopathological examination using periodic acid-Schiff staining of nail clippings” in a recent issue of IJDVL.[1] The authors’ findings about high sensitivity of periodic acid-Schiff (PAS) staining in the diagnosis of onychomycosis is consistent with previous studies on the subject.[2,3] However, certain fallacies of PAS staining in this context deserve consideration as appended below.

Comparison of potassium hydroxide mount and mycological culture with histopathologic examination using periodic acid-Schiff staining of the nail clippings in the diagnosis of onychomycosis.

BACKGROUND Onychomycosis is a common problem noticed in clinical practice. Currently available standard laboratory methods show inconsistent sensitivity; hence there is a need for newer methods of detection. AIMS This study involves comparison of standard laboratory tests in the diagnosis of onychomycosis, namely, potassium hydroxide mount (KOH mount) and mycological culture, with histopathol...

Comparative study for the reliability of potassium hydroxide mount versus nail clipping biopsy in diagnosis of onychomycosis

Introduction: The clinical pictures of nail dystrophies caused by different fungi are seldom diagnostic. Mycological confirmation with appropriate diagnostic techniques including direct microscopy and fungal culture is important to ensure correct diagnosis and treatment. Microscopy can be performed using potassium hydroxide (KOH) mount and histological evaluation of nail plate specimens with th...

Onychomycosis: Current trends in diagnosis and treatment.

Onychomycosis is a fungal infection of the nails that causes discoloration, thickening, and separation from the nail bed. Onychomycosis occurs in 10% of the general population, 20% of persons older than 60 years, and 50% of those older than 70 years. It is caused by a variety of organisms, but most cases are caused by dermatophytes. Accurate diagnosis involves physical and microscopic examinati...

Polymerase Chain Reaction-Restriction Fragment Length Polymorphism as a Confirmatory Test for Onychomycosis

BACKGROUND Onychomycosis is a fungal infection of one or more units of the nail caused by dermatophytes, or mould and nondermatophytes yeast. Investigations are needed to establish the diagnosis of onychomycosis before starting treatment. Several investigations methods for diagnosing onychomycosis are microscopic examination with 20% KOH, fungal culture, histopathology examination with PAS stai...

Lymphoid Leucosis and Coligranoluma in a Budgerigar (Melopsittacus undulatus)

An adult female budgerigar (Melopsittacus undulatus) presented with abdominal enlargement. The condition of the bird deteriorated after needle aspiration for cytological examination. The budgerigar was euthanatized and a complete necropsy was performed. Microscopic sections were prepared and stained with hematoxylin and eosin, Gram staining, periodic acid-Schiff (PAS) and acid-fast staining. Es...
